      针对纸基底上显色反应的检测,提出了一种基于光热-激光背向散射干涉的纸基生物分子定量检测技术,并进行了实验验证。该技术通过泵浦光激发纸基底上有色物质的光热效应,产生的热量传递给毛细管,并引起毛细管内液体折射率变化,从而使得探测光的背向散射干涉条纹发生移动,移动量与纸基底上的有色物质的浓度正相关。文中以肾病生物标志物视黄醇结合蛋白(Retinol Binding Protein , RBP)为例,利用酶联免疫吸附测定(ELISA)方法使得RBP显色,并将显色溶液滴加至滤纸基底,采用泵浦光调制的光热-激光背向散射干涉技术,实现了RBP的定量检测,检测限为0.22 μg/mL。

      A quantitative detection technique based on photothermal laser backscatter interference and filter paper substrates was proposed for the detection of color reactions on filter paper substrates, and experimental verification was conducted. The photothermal effect of colored substances on the filter paper substrate was excited by pump laser, the heat generated from the filter paper substrate is transferred to the capillary and induce a change in the refractive index of the liquid inside the capillary, leading to the shift of the backscattering interference fringes of the probe laser, and the amount of movement is positively correlated with the concentration of colored substances on the filter paper substrate. Taking the renal disease marker Retinol Binding Protein (RBP) as an example in the work, enzyme-linked immunosorbent assay (ELISA) was used to color RBP, and the color solution was dropped onto a filter paper substrate. A pump laser-modulated photothermal laser backscatter interference technique was used to achieve quantitative detection of RBP with a detection limit of 0.22 μg/mL.
